Nuclei isolation from mouse brain tissue - Nuclei isolation (Jun/29/2006 )

Hallo everybody,
I try to isolate nuclear RNA from mouse brain tissue(hypothalamus). Since now, my results are very sad: I have a lot of genomic DNA contamination...does anybody have experience with nuclear RNA isolation from brain tissue? I think it would already be fine to have a method to isolate the nuclei properly...I would be thankful for any help, Susanne

I'm not familiar with nuclear RNA specifically, but you can get around the problem by DNasing your sample. Why is total RNA not acceptable or what is your use for the RNA?

I think you are correct and a nuclear extraction and then a RNA isolation will target the compartment specific RNAs, but you will still have the genomic contamination.
