which voltage for which agarose concentration? - assuming same size of gel, how should voltage be adapted to agarose concentratio (Jun/28/2006 )

Can a higher percentrage agarose gel be run at higher voltage than a lower percentage gel? Or does the increased density cause higher resistance, more warming, and therefore requires lower voltage? huh.gif

Thanks. smile.gif

The gel is essentially "invisible" to the electric field -- what matters is the buffer conductivity. So, there should be little difference between gels of different percentages in terms of heating.

Thanks. So, higher voltage for TBE which has lower conductivity and thus heats up less. While TAE has to be run at lower voltage. Correct?

Exactly.
