how can i preserve the purified mAb for a long time? - (Jun/25/2006 )
i purified the monoclonal antibody from mouse ascite by protein A method,but preserve is a problem, i will appreciate your suggestion,thanks!
As good as I know it's better to keep antibodies in rather high concentration (~10mg/ml) with addition of ~1% BSA for stabilization and sodium azide or merthiolate as preservative (search web for specific concentration of those agents). Keep at +4deg.C for days/weeks, at -20deg.C for months/years or at < -70deg.C for years.
PS. If you want to concentrate antibodies by ultrafiltration, some say that stirred cell is not recommended, use centrifuge tubes (like Millipore Amicon/Centricon) or hollow fiber.

the aliquot in use should definately contain sodium azide 0,05% and be kept at 4°C
I agree with kylvalda. I even reuse Ab diluted in milk if I add 0,05% sodiun azide to the milk up to 2 months
Our everyday use antibody aliquot is in -20C and others stored in -80C. Although I know people in some other labs store antibodies (in current use) in 4C for upto a month.

i first do this experiment,and meet with lots of questions. and after purified by protein A method ,i found the sensitivity decreased a lot. so i worried about the preservation,and 10% glycerol is one of the methods from references,just want to know more selections.

if you use mab everyday u can aliquot it in many smaller fractions ,a fraction in 30% glycerol PBS in -20 deg C for erveryday use, other fractins u can store no add glyerol in -80 deg C. other thing dot not dilute mab ,high density >1mg/ml is good ideas.
