gsamsa - Gene array vs PCR (Jun/22/2006 )
Hi.
I have no knowledge at all of gene array, so I hope someone here could help me.
I've done PCR of a sequence which according to a published gene array data from a university should be present. But after several attempts, improving conditions and so on, I cann't get anything.
Up to what point I can trust the gene array data? I'm using a cell line which comes from bone and the gene array data refers only to bone.
How should I interpret my PCR results in contrast to this gene array data?
Thanks in advance.
:-(
Correct me if am wrong.. but the results of an array can only be validated (in this case by PCR) in the same initial samples unless they are very related.
Am also tryng to validate some data of an array first by wester, (no results) then by real time PCR (more convincing)

Do you have a positive control in which you see an amplification?
Microarray data does not necessarily true. That's why it require validation.
