adding antibiotics to cho cell media - (Jun/14/2006 )

i am growing cho cells and have been having contamination problems for the past week...i am going to be making new media and i have heard that many people add penicillin and streptomycin (penstrep) and i wanted to find out if there are any disadvantages to using the penstrep?.....i will be using these cells for stable cloning and didnt know if there was a negative in using the penstrep and then switching to high g418 media?

Hi,

I always use PEN/STREP in the cell culture medium. Moreover, when I work with stable clones, I have PEN/STREP and G418 in the medium.

Usually, one does't use antibiotics only during transfection because of transfection inhibition and/or cell death.
