Macrophage and IFN - Macrophage's activation (Jun/06/2006 )

Hi everybody,

I am working on bacterial infection in macrophage (Mf4/4 and J774) and I would like to compare the effect of activated macrophage compare to normal macrophage during the infection. Do you know the concentration of IFNgamma I need to use and the protocol of activation?

Dear Biofred,

We have used 50 units/ml IFN to activate macrophages like J774 and Raw 267.4, but you get a dose dependent activation upto 1000units/ml. You also get a synergistic activation if you add LPS. Maximum iNOS activation is seen at 10ug/ml LPS and 50units/ml IFN gamma.
