TB1 Competenet Cell - (Jun/02/2006 )

Dear Fellows

I have a problem with TB1 cell. I cloned my gene in pMALc2x and tranformed into TB1 cell and the cells grow well on amiciline palte when i induce lb media for overnight grwoth the cell doesnot grwo and grow very slowly and i leave it for 48 hr but it does not gorw at all and in one culture i have growth but when i extract plasmid and run it on agrose gel i get smear and no bad at all.
what it can be a problem.
please suggest me.

U might be getting the smear bcoz of some enzyme which is also being extracted during plasmid extrcation which is digesting the DNA.

It is possible that the overnight LB cultures grows slowly, this can happen due to the plasmid.

So whats its solution and how can i get rid of it
