Quantification of heme proteins? - How to? (May/31/2006 )
Hi everyone,
at the moment I'm trying to quantifiy a heme containing protein. this is not straight forward, as it seems that tests like Bradford and Lowry are disturbed by the heme, so lead to wrong concentrations. any suggestions would be very welcome.

you could try reading the heme directly. i don't remember the wavelength (i think it is somewhere around 450nm) but you can find out with a scanning spectrophotometer.

yep, but then you have to be sure about the oxidation state. another problem is that the absorption of the heme changes due to the enviroment, so it's different in protein A and B
