Calcium Imaging - (May/19/2006 )

Anyone worked with calcium green? Do you know if it is cytosoli Calcium what we measure and not RE or mitochondrial? by confocal I have high levels of Ca in nucleus compared o cytoplasm.
I also wanted to determine calcium by FACS to see movements after stimulation, but I think might not be the best Idea since calcium might change in miliseconds while incubating with the indicator.
Any suggestions please?

i used Fluo-3 for this kind of experiment and it works well in real-time aquisition by flow cytometer even at RT wink.gif
