dissociation of spheroids - (May/11/2006 )

hi.
I am trying to count the cellnumbers of cells growing in neurospheres. While dissociating them with trypsin (and by now also accutase, EDTA, dispase) I always end up with the problem, that the cells form "slimy clumps". the cells seem to have little hooks with witch they stick to each other.
Has anybody perhaps observed the same, and even better has an idea how to circumvent the problem and end up with nice single cells???

Have you tried treating them with dispase?

I am also working with spheroids. I get them nicely in suspension by adding accutase.
