medium must be changed after Calcium transfection? - (May/10/2006 )

Hi, guys,

I need to do calcium phosphate transfection on 293T cells. My questions is: is it a must to change the medium (~20 hrs) after transfection?

I have left the medium on for more than 24 hrs. but then it depends what will you do with the transfected cells. If u r going to produce virus, then u want to change the medium.

I just want to express GABA A receptor (a membrane receptor) and harvest them 60 hrs after transfection.

I guess it might not make a difference. But the medium might turn yellowish by 60 hrs and the cells might not be healthy. So u could change the media after 20 hrs and then wait for 40 hrs and collect the cells.

pH is case sensitive but maybe also the salot concentration may affect the cells.
