isolation of phage dna - isolation of phage dna (May/10/2006 )
i am trying to purify ctx phage from V. cholerae, but i an getting a lot of chromosomal dna contamination..
can anybody help and suggest something

Phages are supposed to be extracellullar most of the time. So they must remain soluble in the media. So if the phage you are trying to purify is extra cellular there is no need for breaking the cell, just collect the medium (and disgard the cells) and do a phenol/chloroform extraction to clean the phage DNA from the phage proteins. Then ethanol precipitation.
