Gel extraction - just wondering!!!! (May/08/2006 )

Hi there all,

Just wondering, im using the Qiagen Gel Extraction Kit, i have extracted the desired band since i do face unspesific binding. Is it ok to keep the extracted gel in 4C overnight and carry on the process the next day????

Hi,
I have tried this out when i got busy with some other work. The gel slice can be very well stored at 4C ( i stored till 24hrs) and it worked fine for me. There was no DNA damage or loss due to this.

Did the same thing too.. kept my gel overnight at 4 degress.. able to use still...

Did the same thing today, and there was no result beyond expectation.

Thanks loads, cause i was really busy today. I extracted the gel at bout 8pm. I'll do the extraction tomorrow morning. I think about 12 hrs different. Hope the result will b the same.

I normally store the gel at -20C for longer period of time, during weekend or for a week or so. There is no loss of DNA.

i agree with scolix. there is no problem storing the gel slice at -20°C. i have done it several times before and there was no lost of dna observed wink.gif

Theoretically the only problem would be the presence of exonucleases "eating" the overhangs from the cut DNA. So I would put it in -20

I kept my gel slice o/n in -20 and used next day w/o any problem!
