sequencing problem - (May/08/2006 )

Hi I have cloned gene of interest and sent to sequence it But after sequencing i saw that 8 bases are abcent.Is it possible to cloned the gene with 8 bases abcent??(8 bases are internal bases)

I think that could be a real problem, if the sequence is accurate. I might repeat the sequencing reaction to confirm...but at any rate, that's a frameshift mutation, to say the least; perhaps your polymerase induced some errors?
