Guanidine-HCl solution storage - (Apr/28/2006 )
Hi, everybody:
I´ve been trying to extract DNA from solid tissue using a 8M solution of guanidine hydrochloride with ammonium acetate or alternatively with sodium acetate, pH 8.
The aim of using this solution is to purify the DNA, removing proteins.
It works most of the times, except when the solution is stored for several days at 4ºC. In this case, no DNA is obtained.
I wonder how this solution should be stored for this not to happen or if it is better to use a fresh one.

i use solutions that contains guanidine hcl.
they are stored at 4°, but before using them, i heat them to 25°.
