phenol-chloroform or just chloroform - (Apr/21/2006 )

Hi, i need to isolate DNA from a virus, so after the lysis what is better extract the samples with phenol-chloroform or can i use just chloroform, and then extract with ether????

both (phenol:chloroform then chloroform: IAA) or chloroform will make do. Don't use phenol only cause it tend to separate less well with aqueous phase and you'll have OD260/OD230 < 2.0.
