Primer dimers?Contaminations? - Unexpected bands! (Apr/20/2006 )
Hi,all.
I have performed PCR for more about 1 month. Now,a new problem emerges.
Thanks you all for any suggestion in advance.
The bands of first row are reference of HGH(human growth hormone),something are good.
The bands of second row are expected fragments.
But,what are the bands of the third row?

The first row should be counted for well.

Try running a ladder with your samples, it helps a lot to know the size of the things we are seeing. Also try running your gel for longer so that there is better separation. Also did you run a negative reaction (just water instead of template), and do the bands show up in it?
At the moment it looks like primer dimers. but can't say for sure.

where is your negative control (without template)?
It should answer the question of pcr primer dimers...

So in negative control you should get primer dimers, right?

No. If you have primer dimers, you will have them in the negative control (where you have no template) as well.
