siRNA delivery into human primary macrophages - (Apr/18/2006 )

Hi!

I would like to silence a gene in human primary macrophages, which are notoriously hard to transfect. Does anyone have experience with tranfecting these cells? Maybe a lentivirus based system is the way to go, but also way more work (and we only need transient silencing). So I am a bit hesitant to go there. Has anyone succesfully tried delivery using a lentivirus system? I would be very happy with a 50% efficiency...

Hi,

We use nucleofection (AMAXA) and will be shifting to siRNA delivery into monocyte derived macrophages from buffy packs. Havent tried it yet, but we are confident it will work. Might be an option for you?

Thanks for the suggestion! I have looked into that and found out that we even have the right machine in the building, so we will try it for sure.

I am using Lenti virus to knockdown a few genes. It works efficiently as long as you have enough viral particles to infect 80% or more cells.
