Needle puller for C. elegans injection - (Apr/12/2006 )
Hi guys,
I'm just starting with some C. elegans injections to make transgenic lines. We have a Campden needle puller but this is set up for mammalian injection needles and the needles are hopeless for worm injection. Does anybody have any ideas about the settings we should use on this machine to pull good (reasonably short and blunt!) needles for worm injection?

I've never done this before, why do you want blunt ones? The gonad is small wouldn't sharp ones be better?

you need long and thin needles for the injection into the worm. possibly closed at the top, you break them during the first injection. try to pull the needle faster or heat a bit longer, to get long tips. the settings depend on the instrument and can very between them (not only between different brands but also actual pieces of equipment).
