CFSE staining - (Feb/03/2001 )

I would like to stain murine T and B lymphocytes with CFSE in order to detect them in vivo by immunofluorescence after iv transfer into host mouse. I would like to know if I should use special procedures to prepare my cryosections (is fixation in acetone OK ? should I fix the tissue cryosections or the spleen sample before embedding it and freezing it at -70 ?) .... Thanks a lot for any advice and suggestion.

Just cut section and see on microscopy, please do not fix the sections. Also, You still can see labebed cells wif section is kept at 4oC.
