Puzzle about methylation on plant microRNA - (Apr/06/2006 )
It seems that plant microRNAs are methylated at 3’, which should be 2’-O- methylation; however such modification should block polyadenylation(polyA tailing) of microRNA by E-PAP or Yeast PAP in vitro.
On another hand, a polyA tailed RT-PCR protocol has been validated for plant microRNAs and there is also a microRNA cloning work based on polyA tailing technique for rice...
So any ideas for this issue?

Are you talking about micro RNAs or messenger RNA? microRNA may not be at or near near a polyA. More than likely they are from non-coding regions.
Of course your messenger RNAs will have the polyA...
