PCR mix and DEPC water - (Mar/29/2006 )

Is there any problem to do PCR mix with DEPC water???
Because I've already heard that DEPC water degrade DNA, is it true??? unsure.gif

DEPC can hinder the activity of RNases. If your water is a commercial brand it is most likely treated to be RNase-free.

I've used it for PCR and have not had issues. If you see a degradation of DNA you have Dnase contamination.

besides, the depc is decomposed by autoclaving after treatment. so there should be no intact depc in the water.
