Amastigote of Leishmania-ELISA - Detaching of bound antibody (Mar/25/2006 )

Does anyone know how bound antibodies to cell surface are removed or detached? Can I use pepsin digestion or buffer with high pH on the parasite cells? I want to use these cells for coating on the bottom of ELISA well and prevent false positive.

don't you have any antigenic peptide to compete with natural Ag expressed on the cells?
heating or high ionic strength (including low pH) are generally used to break Ab-Ag interactions in biochemical assays, but Ag will be denatured and no more recognizable in subsequent ELISA...
