How to know if a protein that has autokinase activity is a kinase? - (Mar/24/2006 )
Hello, I would like to know if a protein with autophosphorylation activity can be a kinase? How can I test this? a kinase assay? if I dont have a specific substrate protein, how can I test wheather or not this protein is a kinase?
Im reading about a Hsp protein that has autokinase activity and I was wondering how could I test if this protein is a kinase.
Any knowledge about this topic will be greatly appreciated.

Most kinases have activity on myelin basic protein (MBP). This is a general substrate available for purchase. There are also peptides that can be used, like the S6 kinase substrate peptide.
Do you know the sequence of this protein? If you do then a simple blast search will tell you whether or not it has homology with other kinases. If you don't have a sequence, I suppose you could try to isolate it and have it sequenced yourself.
