How would we know that the number of cells is the same when you aliquot them into 12 well plates?

First, you do a cell count of a given volume.

Then you dilute this so that you get a given number of cells into the volume you want to plate and then you mix it all and plate your diluted cells. You will never get 100% exactly the same number of cells, but approximately...
