hypoxic cells - how to make hypoxic cells??? (Mar/20/2006 )

How can we make the cancer cells hypoxic in order to determine the effect of drugs, radiation or other stresses??? Should we have to grow the cells in hypoxia chamber for a long time or 1 hr or few hours are enough so that the cells behave as hypoxic cells???

You can use a hypoxic chamber or an incubator where you can reduce the O2 content to 5%.

The time is dependent on what you want to measure. Early-response genes get switched on within 30-60 min. Normal genes require ~8-24h. Protein changes will need > 8-24h.

Thanx a lot.
I want to see the effect of radiation in the expression of some enzymes,p53,HIF and IL-8 in hypoxic cells in different time intervals like 8hrs,24 hrs and 72 hrs. So how long should I incubate my normal cancer cells in the hypoxia incubator before radiation treatment so that my results are at least comparable to in vivo condition. Is that 1 hr incubation only then radiation or after 8 or 12 or 24 or 72 hrs???What is your suggestions in this matter.
