Hello folks.
Speaking of RNA extraction/handling is there anybody who does not use this DEPC treatment?
Is there any substitute for DEPC to preserve RNA integrity in water solution?

i don't know if RNA later is ok for that. But you need also to take in count that anything which is added in the storing-solution may interfers with subsequent reactions. As DEPC is well inactivated by good autoclaving, it' the most used.
I keep my RNA in deionized formamide. But if i would do a RT PCR, i would have to pellet them and resuspend in xater, as formamide is not ok to do RTPCR.
So if you think to use an other storing solution, remind this example to see it's possible with what you aim to do.

Actually, what I meant to check with other people is the question whether
some folks run their RNA business using just deinozied well-autoclaved
water. I know at least couple of people in our isntitution that do this
on regular basis. And they claim they never have problem with RNA integrity.
Any idea?

well... RNAses are sterilized by autoclaving. So except if the water is very pure (i consider my miliQ not RNAse pure, so for this purpose, i would buy molecular water if i don't want DMSO...) i use DMSO treated one.

We use water straight from our milliQ (synthesis) machine for all RNA and PCR work. I'm never confident that DEPC is completely removed, and I have experienced problems downstream when using it
