Fred, thank you for your reply. Since as you suggested i am having my band of interest I am not worrying that i dont have my protein. Missense mutations are my worry but till now i didnt try anything to varify that. My update: I have successfully solubilized my protein with Sarkosyl....but now it is not attaching to the beads properly. On the other forum ive been suggessted to optimize how much excatly of solubilizing agent I need....so now im working on that. thank you very much for your help, guys.
