extraction RNA from coronary artery endothelial cells - yield problem (Mar/14/2006 )

hi i get very low yield of RNA ising RNeasy mini kit
only around 10ug from 4-5 T-75 flasks
Is there any way to induce RNA levels befoer extraction to get better yields of RNA

how many cells do you have in your flasks?
for problem of yield, i think first step to check is the proper amount of loading solution you use (freshness too).
In elution, the pH is also case sensitive. A pH close to 8 is good and i was surprised that quite little changes of pH (i mean i thought 7 would be ok too) can drop the yield to ground level...
