Combine fluorescense microscopy with HE-staining? - (Feb/24/2006 )

Hi,

I do fluorescense microscopy with monoklonal AB, Cy3 secondary AB, on tissue sections. Now I want to stain the hole slide to see the structure. I want to do that Haematoxilin Eosin staining. Is it possilble to combine the 2 methods, fluorescense and HE-Staining, or is there another staining method, that works better with fluorescense?

since you can obtain a picture of your HE staining in the bright field channel, you would be able to merge others channels like fluorescence on it...

Hematoxylin is autofluorescent and eosin may be (I can't remember well about that one) so I think it's a bad idea to do both on the same section. I suggest you do as most people do which is use a nuclear fluorescent counterstain such as DAPI or Hoechst 33342 for overall structure or worse comes to worse take a phase contrast picture and do overlay.
