MCF-7 Cell line - (Feb/15/2006 )

I am having trouble in maintaining MCF-7 cell line. After few passages (2-3) cells start degenrating with most of cells in suspension and also change in morphology. I use MEM from ATCC supplemented with 10%FBS and insulin. Does anybody has idea how I can maintain the cell line and also does adding 10nM estrogen will help?

Apart from just insulin I add sodium pyruvate, non essential amino acids and glutamine and other good stuff to my MCF-7 media. I also use the ATCC MEM and there had been a slight chagne in the morphology but it is fine (knock on wood

Also addition of E2 will help in increasing the noof cells. You can try adding 1-4 nM of E2.

The ATCC MEM medium already contains Sod. pyruvate (1.0 nM), non essential amino acids (0.1 nM) and sod. bicarbonate (1.5 g/L). Do I need to add more of it?

I grow mine in DMEM with 10% FBS and EGF. They grow very well.
