help with background in northern blot - (Jan/26/2006 )

Hi! I'm doing several northern blots, but i've got high background with one of my probes. I can see some signal but this background is very intense and I can't observe if there are some differences between the samples. I've tried to dilute the probe, but I continue getting this high background. What can I do?. Thanks for all

you can get rid of backround by autoclaving or at least by filter sterilizing your buffer used in washing steps. you should also ıncrease the amount of hybridization buffer. the main reason of the backround is the insufficient hybrıdızation buffer.
