bubbles in SDS-PAGE - (Jan/26/2006 )
Hello there,
I have a little question :
It has been years I've been pouring SDS-PAGE without any problems, and today something strange happened.
As usually, I prepared the gel yesterday evening, and after one hour polymerization I put it in the fridge. this morning, there is a bubble in the gel,and while it was getting warmer in the lab, the bubble spread on all the width of the gel.
I guess I cannot use it anymore.
I do not filtrate it neither de-gaz, but I 've never done that before. do you think this is the problem?

It it not bubble in the gel, it is just the bubble between the gel and the glass plate, as the gel contract in fridge and expand (a little) in room temp.
There is no problem in running this gel. Just be careful when handling as the plate may fall off from the gel easily.

I ran the gel, and it's fine.
I was so surprise, it's the first time for 5 years I see that.
There is always a first time !
(However, I also experimented it while I let the gel at room temperature ?!)
