Extracting DNA from serum - (Jan/20/2006 )
Hi i need some help:
i have been given the task of finding a way of extracting DNA from sera without using a qiagen kit. Does anyone know of any protocols or papers relating to this?
Thanks
do you mean without a QIAgen kit, or without any commercial kit (ie using homebrew buffers)?
Without any commercial kit.
thanks
Hi bsuac6,
Maybe it's not the safest protocol but I've always used phenol-chloroform for DNA purification from Serum and Plasma and always obtained better yields than with any other commercial kit (I'm not very fond of them, I have to say... ). Maybe purity is not as good but I've detected very small quantities of DNA using PCR after DNA purification.
It has always worked for me...

I agree with Esther. Phenol/chloroform is and always has been my favourite method for isolating DNA.
Thanks guys, i tend to use phenol/chloroform with whole blood, so i guess i might as well use it for sera.
Thanks again.

Hi,
I have:
od 260 od 270 od 280
0,066 0,103 0,044
What's the meant of these data?
I know that there is protein contamination, but I don't know the meant of this od 270....
I used 2 cycles of PCI extraction and 1 cycle of microcon 100......

Thanks!
