DGGE gel problem - (Jan/13/2006 )

Hi
If anyone can solve this, I will be very happy.
In the DGGE gel which i try to run, the dye after loading it just moved a lit bit but failed to move further.
I used a gradient of 40 to 60% and 8% polyacrylamide gel.

The voltage i started with 100V and later reduced to 70V. After running overnight still the problem existed.
When i stained my gel i didnot see any DNA.
Please help me to resolve this.

Hi there,

sorry for seemingly asking a very dump question, but did you check that your upper buffer reservoir was still full? Sometimes it leaks and if there is no buffer there, there's not going to be any current running through... alternatively, if it is a ready-made gel you bought from a company, you will have to take away the little plastic strip that covers the lower reservoir access to the gel.
