RNA extraction from formalin fixed paraffin embedded tissue - protocol? (Jan/11/2006 )

Hi,
I would like to extract RNA from formalin fixed paraffin embedded tissue. Does anybody have a good well working protocol for this? I would be very gratefull if somebody could share me his/her wisdom concerning this!

Hi Karolina,

Although you can get some RNA from formalin fixed and paraffin embedded tissue, it will be mostly degradated. I doubt you will any meaningful expression data from it. So I would say forget about it. One of my colleagues tried to do this and use the RNA for hot RT-PCR. At the begining, I opposed to his idea, but he insisted on carrying on his plan. He ended up with nothing after wasting months of time and hundreds of $$$.
