KB8301 MMP protease inhibitor - (Jan/06/2006 )

I've just been informed that the MMP inhibitor KB8301 is no longer available commercially. The company that holds the patent withdrew the product, presumably because they found some thereputic use for it. It is the only inhibitor I know that effectivly prevents the shedding of Fas ligand from the membrain surface. Are there any fas people out there that know of another inhibitor that works, or where I can get my hands on the precious KB8301. Thanks

No, I spoke with a guy from pharmingen (That is where we used to get it) and they have no replacement. I contacted Kenobo Ltd in Japan, they refered me to another company, who said they no longer have the compound . Dead end, I have about 5ul left. It hasn't been a pressing issue for me yet, but I do have my eyes and ears open and will let you know if I find something

I work on Fas Ligand too! I have tried long and hard to find KB8301 which BD no longer produces. One alternative to try is GM6001 from Chemicon, although it doesn't inhibit MMP7 which, in vitro seems to cleave FasL quite readily. Another to try is MMP inhibitor II from Calbiochem. I have never used either of them and seen increases in surface expression by flow cytometry but I don't really have a very good system yet. They seem to work ok for other people though... (grumble, grumble).

PS where are you guys from? maybe we should stay in touch for troubleshooting/adivce etc....it's always good to have inside info...

PS when did you last get KB8301 anyways??? I was under the impression that it had been off the market for a really long time.

I'm not sure when we last got it, I've only been working on this project for a year and it was in the freezer when I got here. I'm new to the fas ligand field and had I known it was in such short supply I would have used it more sparingly

Thanks Jon and mountainman! Anyway, I'll give MMP inhibitor II a try. I know that BB94 which was used for the original FasL shedding research is no longer available too.

Mountainman, do you know how long pretreatment is needed for MMP inhibitor II to work? Thanks!

So did u guys find the FASL inhibitor KB8301? I was also looking to purchase to this FASL inhibitor, but I didn't find with well established companies.
Jinshu how was your impression with regard to MMPII, as u said u will use it. Is it good to use as per ur experience? Or u found any other good alternative?
