Now I have research about siRNA in meristem and primordial leaf, I would like to investigate siRNA in primordial leaf, but primordail leaf is very small region. I would like to know how to extract siRNA from primordial leaf and How to detect siRNA in primordial leaf? Please tell me...

You could try Invitrogen's charge switch technology for nucleic acid extraction, it is supposed to be able to extract DNA or RNA from as little as 1 cell. It is probably pretty expensive though.
