membrane protein extraction from dissected thymus - (Dec/28/2005 )

Hi, everyone

I'm working glycosylation alterations during the embryogenesis. But I don't know how can I extract membrane proteins from thymic tissues for WB? thanks in advance. merry Christmas

Are you looking to isloate the membrane proteins from the rest of the cellular proteins? If so you will need to look into membrane isolation protocols. Once you isolate the membranes you want to look at just simply dissolving them in SDS containing sample buffer should be sufficient to run the membrane proteins on a gel for WB. If the gel bands look wavy, you can double the concentration of SDS in normal sample buffer (3% --> 6%) to help dissolve all the lipids.
