maxiprep on SURE strain - (Dec/23/2005 )
Recently, I try to maxiprep a plasmid in SURE competent strain (Stratagene). I tried 2 times just like what I usually do in DH5a, but only get very low yield plasmid. Does anyone know if there is something special when deal with SURE cell in maxiprep?
Some strains (I haven't worked with SURE) have other growth characteristics than others. Using SURE cells, I assume you're having repeats in your DNA sequence? For this purpose I use Stbl cells and grow them several hours more than I usually do with DH5alpha cells, because the stbl cells grow more slowly (so maybe for SURE cells it's the same deal?). I go up to 24 hrs (on 30°C with 180 rpm rotation speed with stbl's and get very high yields with maxi preps). Another thing you might wanna try is not growing them in LB but for instance in terrific broth, which could give higher yields of plasmid as well.
