I try to obtain good splicing extracts from yeast, but I don't achieve it. Could anybody tell me if there is a very critic step on the protocol from extracts preparation?

We use the protocol of mortar with nitrogen (Umen & Guthrie protocol).
I think that's very easy, but our eficiency is low, sometimes we can see the intermediates (and very, very few) but not always.
