luciferase reporter vector - promega (Dec/04/2005 )
Hi
I am beginning to do luciferase assays for my promoter and using the promtoerless basic pGL3 luciferase from promega
What other control vectors do I need to use. There are some Tk-Renilla vectors and CMV-Renilla vectors . How do I decide which one I use. I am studying the promoter of a human gene. Does that affect the choice of the control vector used.

Tk and CMV are different promoters, they drive the expression of the luciferease. Tk is, from memory, weaker thean CMV. So, if you've got time, you could compare the two, to see what strength you need in order to see a result.
