Whats the best media to get the fastest E.coli growth at a low cost? (only conce - (Dec/01/2005 )

I dont need to isolate plasmids, or produce a protein.. I just need to produce the highest cell biomass of E. coli possible in 8 hours. I start with a 200mL shakeflask inoculum, then add the culture to a 9.8L fermenter (10L total). The fermentation will occur in a total of 8 hours..

What media would you recommend to get the highest cell biomass? Right now im thinking LB (1% tryptone, 0.5% yeast extract, 0.5% NaCl) but surely theres a better alternative out there.

Id use superbroth (3.5/2/0.5% of those 3 ingredients respectively) but its too expensive, same with terrific broth (1.2/2.4/phosphates).

Any suggestions? Should I add phosphates to my media? (KH2PO4/K2HPO4)? Any other component suggestion that I should use? (soy-peptone, glucose, lactose, etc) Should I go with the Glucose M9Y media? Or should I use some wierd % of trpytone/yeast extract/nacl like 1.4/0.8/0.7%)

You should use Mach1 cells (Invitrogen). They grow very fast compared to most other coli strains (they are not K-12). Aeration is critical. Use baffled shaker flasks or good bubblers. You might think about an oxygen enriched atmosphere.

couldn't you possibly make the superbroth yourself? or terrific broth, for that matter. the recipes are pretty simple. i purify protein a lot, and make my own superbroth.

Adding some glycerol used to double the cell density, for over night cultures, but I do not know if eight hour would be enough to get the full effect, and I have only used it in 1-3 ml cultures in tubes, never in larger volumes.
And, I don't remember the name of the broth, or the recipe. It probably were a recipe once, but over the years it developed into - throw a little glycerol into the tube if hunch told me I could use some extra cells.
