ELISA versus ELISpot - (Nov/28/2005 )

Hi!!!


Is anyone can say me what is the major difference between an ELISA and an ELISpot. Is it only the method of revelaltion?

With an ELISpot you can detect the emission of cytokines by T cells of others but it is possible to detect emission of cytokines by the method of ELISA, the only diffrence reside in the secondary antibody which is coupled to HRP or something else... Is it correct??

And if so, ELISpot is used because more economic ?

ELISA gives you information about how much cytokine is produced and secreted at a given point of time, but no information about how many cells are secretors of the cytokine.

ELIspot gives you information about how many cells secrete the cytokine, and a fair impression of how much cytokine these cells secreted, but no quantitaive information about cytokine concentrations.

so, those two are not the same and will yield different information, but complement each other rather well...

Detection of intracellular cytokine by FACS may be an alternative to ELIspot, since you get information about how many cells are producing your cytokine, and depending on the florescence level, how much cytokine is produced in every cell (-though, no quantitaive information here, too)
