No DNA on gel, but get PCR product - (Nov/23/2005 )
Hi,
we try to extract DNA from Drosophila and we get DNA cocentration on spectrophotometer 8,9
and ratio 1,9. We try to run this result DNA on agarose gel 0,8 % but there is no DNA.
we try to use it on PCR for RAPD and Strange......we get band from this PCR.
what happen to DNA sample?
thank,

whats the predicted concentration by spec? And also, how much did you load on a gel? I am not surprised you got a product by PCR..you can dilute chromosomal DNA up too 100, 000 times and still get a PCR product. You may not be able to see it on the gel because the concentration is very low.

There is nothing wrong with your DNA sample. It's just too low a concentration to see on your gel.
