Slow freezing and thawing of FCS - (Nov/21/2005 )

What is the effect of multiple freezing and thawing on the Foetal Calf serum properties' cell culture nutrient??
any ideas...anybody????

One theory is that the growth factors and other important nutrients within the serum will degrade faster if the serum is freeze-thawed regularly. I would recommend derosting the serum overnight at 4˚C, heat-inactivate if required (56˚C 20 min) and then aliquot into 50 ml lots and store frozen. When required, defrost the alquot, dilute in the media bottle and use as required.
