Primary culture of breast tumour - Does anyone know the tissue slice method? (Nov/17/2005 )
Hello everyone,
I am trying to establish primary cultures from breast tumour, and have been using the explant culture method so far, ie. just mincing with microdissecting forceps and scissors and culturing the mince right away.
However, there are comments that mechanical/enzymatic disaggregation disrupts the normal tissue architecture and cell-cell communications that exists in vivo
The tissue slice method apparently maintains the tissue architecture. Reference is
Hood CJ, Parham DM.
A simple method of tumour culture.
Pathol Res Pract. 1998;194(3):177-81.
PMID: 9587936 [PubMed - indexed for MEDLINE]
but I can't access the paper cos my uni library doesn't suscribe to this journal.
Has anyone here tried this method, and if yes, would it be possible for you to share your protocol here? As well as give your comments, tips, etc about this?
Many thanks. Looking forward to some pointers.
Cheers,
labchick.

Sorry, My uni library only have on-line access of this journal from volume 194 (2001).
I think You may have to go to another university's library to find this article.

Thanks Minnie Mouse.
Hmm, does anyone know/practice the protocol anyway? Would appreciate it if you could share it here
Cheerios!
Does your uni not have an inter library loans system where you can order articles to be sent to you from another library?
