Nuclear and cytoplamic markers - (Nov/16/2005 )
Hi everyone,
I would like to know what proteins would be good nuclear and cytoplasmic marker? I am extracting nuclear and cytoplasmic fractions and I want to make sure there is not contaminations.
Any suggestions would be good!

Histone H1 will be a good marker for nuclear fraction. For cytoplasmic fraction GAPDH will work well

or: tubulin for cytoplasmic, myc for nuclear..

I have been using RCC1 (stressgen) as a nuclear marker, it works very well. As for the ctoplasmic marker I have been using gamma-tubulin, but there seems to be some in my nuclear fraction, esp when I fractionate using the Pierce kit. Does anyone have any suggestions about other cytoplasmic markers (I'll try GAPDH), but are other tubulins better? Also, what protocols are people using for fractionation?
