Hi Shashi,

The best thing I've tried for high GC content (70%+) is using Betaine from Sigma, it is cheap and it is what most of the commercial additives are anyway (inc. Q solution). You can pay 100s of dollars for Q solution or about 10 for Betaine which does the same job.

I use Pfu, DMSO, and a lot of MgSO4. my lovely intron won't work with anything else.
