amplification fails to reach log linear phase - (Nov/02/2005 )

We have been trying to quantitate mRNA obtained from single cells. Using these templates, we have been able to amplify GAPDH (but only at 1-20 ng) but not any of our target genes. There seems to be amplifcation but it never reaches log linear phase. Any suggestions as to what we can try?

What cycle number are you getting amplification? What do your curves looks like?
